Do people ever contact a business asking if they're looking for help? Is there a way to send your resume to a company without sounding like a pathetic desperate loser? I'm considering a career change and there are a few companies I'm pretty interested in that I don't think are hiring, and I'm not sure if theres a tactful way to get my resume to them in case they end up looking for someone in the future.

I'd grab an annual reportof the company first and look for a reason you think you would like to work there. A bit of advance work and if you get an interview you have extra details about the company you can throw out then.
I haven't done that myself but I interviewed a new college grad looking for a position in equites trading at my former firm. I did give him an A for planning ahead. His qualifications and personality got him the job though.
Direct answer: Use anything you can to get an advantage.
Do you know anybody who works for these companies? (or someone who knows someone. LinkedIn is good for discovering these type of connections) If so, you could request some informational interviews.
ETA: I wasn't finished with my thought, Nest. Since you're a career changer, informationals give you a chance to not come off as desperate for a job now-now-now, and position it more as "I'm interested in making a change, and I'm doing my due diligence and research to understand what the work is actually like and what it takes to succeed at company X, so I can take the steps necessary to make myself a competitive candidate when the time comes." If you make a good connection and impress in the informational, perhaps it can turn into a referral when there's an opening.
