Here is the background info...
I'm an part time employee who has been getting full time hours since January. I work out of one hospital but get sent over to the 'smaller hospital' usually twice a week. There is only one other pharmacist there. She works the other 2 days a week and then a whole pool of us work weekends. She can never keep up with the workload and I have to pick up the slack the days that I'm out there.
Canada Day (July 1st) falls on a Friday this year and I have the whole weekend off! I'm excited to have a long weekend.
Today, I get a call from the other pharmacist. She is working the long weekend and she wants me to work Saturday July 2nd because otherwise she will be working 5 days in a row and she's "super-stressed out" and has no idea when she will be able to pack for her trip to Europe.
I told her I would look into it. WWYD?
A - tell her no. This is not your problem. Why can't she pack after work?
B - tell her yes, don't be a biotch. So what if your 3 day weekend gets turned into 1 day off, work, 1 day off

C - I think it really depends on her attitude and the culture of where you work. If she is the type of person who would trade days with you when you are in a bind or just because you want to, then I would take the Saturday. If not, I might not. I'm generally not the tit-for-tat kinda person, but when it comes to work, it really depends on the culture of where you work.
For instance, at my old job, if I wanted to go on vacation, no one would cover my work for me. This would result in my working at least half days every day when I was on vacation. Usually, I would get up super early, try to work for about 4 hours, and then enjoy the rest of the day.
At my new job, everyone covers for everyone else if someone needs it, regardless of the reason. On my recent vacation, I did no work, except to respond to a few emails a day (maybe a total of 15 minutes).
So if your workplace is more like my old job, then I would go with choice A, and if it's more like my new job, I would go with choice B.

Honestly, I'd go with A, and I don't think it's being a biotch at all. You deserve a vacation just as much as she does. The scheduling stars lined up for you, and I'd take full advantage of that.
The only way I'd say yes is if you two have a give and take relationship where you both cover each other. If you don't mind giving up your time, and it's not a big deal to you, then, you know, do what you need to do. Otherwise, smile, tell her no this time, but if she needs help getting organized so she can pack quickly and efficiently, you could help with that. (Seriously, I don't think you need a day off before your vacation to pack for your vacation.)

blender - yeah, I have been working crazy stretches (up to 12 days in a row). She works 3 days at our hospital and 1 day a week elsewhere. I get that she likes part time, but when it's your weekend, you have to work extra.
With regards to the culture of this workplace...I've not had to ask anyone to switch shifts because I always book off the time I need ahead of time. For example, by the 20th of this month, I need to book off any days I need off for July. This is usually plenty of time to organize.

Then I feel guilty. It's infuriating because this happens.all.the.time and it makes me feel horribly guilty for not being able to help but I don't want to sacrifice myself.

Ditto the others that if she's covered for you in a bind before, maybe I'd do her the favour. But based on your follow-up, it doesn't really sound like you owe her that.

In agreement with this. She has plenty of time to plan ahead and pack. For that matter, she could do most of her packing prior to that 5 day stretch. I would also take the passive approach and say I had other commitments.

I really do want to go away that weekend since we might need to adjust some of our other plans for the summer due to trying to sell our house/move. I'm glad you guys don't think it's biotchy! However, I am going to feel guilty - I always do when I can't help.
