The night before. Probably an hour or two before I go to bed. Or the day of if it's a late flight! V is much better and always nags me to start packing.
Of course, if there are supplies to be purchased for the trip, I do start thinking about those a few days ahead.

I make a list a few days before that has what clothes I want to bring, any electronics and chargers, all the bathroom stuff. Then when I go to back the night before I can mark everything off and know I am not forgetting anything.
I also like to back light and layer so I like to know what clothes I'm bringing to plan outfits and make my clothes have multiple purposes. I'm a very serious packer.
I make a list about a week before we leave so that i can make sure that if I need to buy anything or wash anything I get that done ahead of time.
About 2-3 days before I leave I'll start putting stuff in a suitcase that I don't use on a daily basis like clothes, shoes, swimsuit, camera, book, etc. Then the night before we leave I pack everything I don't need the morning/day we leave. Lastly right before we leave I pack my daily stuff like makeup, toothbrush, hairbrush, straightener etc.
It totally depends on the trip, but I agree with Eva--I'm a serious packer.
Weekend trip: 15 minute packing. I basically take 15 minutes and throw everything into the suitcase (neatly and organized of course), and if I forget it, oh well, I can always buy it. I used to take forever to pack, and be a serious overpacker. This rule has really helped me cut back.
Longer trip: I start planning a week or so ahead of time to make sure I have all the toiletries and other things I need. I also decide whether I want to go with black or brown clothing scheme (otherwise I pack too many shoes). From there I decide on outfits that layer well. The weekend before I pack everything and make a list of things that didn't make it into the suitcase and then cross them off as they get put in.
