If someone in a cube next to you was continually having loud, lengthy meetings in their cube involving multiple people (there are other spaces available), and the noise was distracting you from doing what you need to do, how would you handle it? Mention it to that person? Mention it to a supervisor? Use headphones and ignore it?

For office happiness, I would go to the supervisor last. I think the easiest fix is to wear headphones. Though, I wouldn't personally do this because I don't think I should have to modify my behavior when someone is being rude. But, with headphones, you don't have to bring it up.
If they are having lengthy meetings, I would ask your cube neighbor to move to a meeting space because it is distracting to you. You could bring it up when she isn't having a meeting. Or, you could wait until they've been there for a while (10 minutes?) and ask if they are going to continue meeting, would they mind relocating.

They're going on an hour now- generally her meetings last up to 2 hours. One of the guys has the loudest voice I've ever heard. As I type, they're talking and cackling over each other and it's driving me crazy- STFU!
You mean you don't throw water balloons over the cube or have a nasty sign outside your cube saying to "Quiet - people are working here".....
I would just ask her to schedule a room when she's going to have a long meeting at her desk, that you can't concentrate, plus you don't want to listen to that crap for over 10 minutes. As I'm sure she wouldn't want to listen to you yap for 2 hrs... head phones are always a good option.
