Recently I applies for an HR Assistant position that was open for my city. I turned my application in by hand and I Think I made a good first impression. Anyway, I was one of the 8 our of 137 applicants called back for an interview. Yesterday I was called to come back for a second interview (3 people were called for a second interview). I really want this job, does anyone have any advice on how to prepare. What kinds of questions may be asked this time around?

Well, I guess it depends on who you're meeting with. My experience is that generally a second interview is with different people than the first. I would be certain to ask questions about the culture of the office, growth opportunities and try to get a feel for whether or not people like their jobs, feel accomplished in them, etc.
If you do get a chance to meet again with the people you met with last time, I would take this as an opportunity to drill down further to really focus on whether or not you're a good "fit" for the company. Ask your potential supervisor about his/her style of management, expectations, etc. Ask them to take you through a "typical day", etc.

I just had a second interview, and I did quite a bit of research on typical second interview practices. I gathered that a second interview will typically cover everything the first interview did, only more in depth.
My second interview consisted of the person who conducted the first interview introducing me to her business partner. The first interviewer explained what we had discussed in the first interview, had me explain some things my self and the business partner asked me questions about some of my answers.
