(I don't know if this has been mentioned on here before, but no one brought it up on the HIMYM post last night.)
Ahem...here is my take on all the little puzzle pieces...
We know that she is Rachel Bilson's roomate and that Rachel was a grad student
We know that she was in that very first class that Ted thought was his classroom
We know that Ted meets her at Barney's wedding
And...wait for it...
We know that Barney has a half sister that is away at college (Barney's dad mentions it when he is over the house for dinner with the family)
So - my guess is that Ted meets Barney's sister at the wedding and they live happliy ever after.
This would tie in all the Uncle Barney/Marshall and Aunt Robin/Lily talk as well.
Now, if I can just figure out who Barney is marrying (and it better not be Robin. Barf!)
Anyone agree? Disagree? Am I a billion years behind bc you all knew this already?

I know this theory was banded around when the show ended last season. It is definitely plausible.
I swear I once heard Ted say Uncle Don to his kids, so maybe Don comes back for Robin? I don't like Robin and Barney either.

Remember the guy from the club that Robin remembers from the mall with the ugly shirt? And he thinks that Robin and Ted get engaged. At the end of that episode he is on the phone and says something along the lines of "I saw that girl again and she is engaged. I guess that is the end of that." And the Ted voice over said - "But it wasn't". I am hoping he makes an apperance again at some point. ( I am willing anyone to be with Robin other then Barney. I just love how geeky he is for Nora.)
I remember a couple people bringing this up last year, I think the sister is a really good possibility.
I remember hearing Uncle Don, too.
I always assumed that all the Aunt Lily/Robin and Uncle Marshall/Barney is just because they're all so close. My parents had friends I used to call aunt/uncle.
I read this theory last season and I think it's a pretty good theory.
I think Barney is marrying Nora. I don't think he is marrying Robin.
I've also heard that theory. I think it could work. I don't know if John Lithgow's character said he had a "daughter away at college" to set the scene for this happening later, or just as a red herring to throw off the audience like they've done with other girls (like the girl Ted bumped into on Saint Patrick's Day).
I don't know why they'd say she's "away at college" if she's in grad school only 40 minutes away, though. That seems like a bit of a stretch to me because it's something you'd normally say about an undergrad living further away. If Barney's sister is really the roommate/Mother, it'd seem more natural to say, "My daughter is living in the city and going to grad school." But it's a possibility that it's her.
I don't want Barney to marry Robin. Robin only seems to get interested in Ted and Barney once other women come into the picture. She was so much more interesting in the beginning when she was an independent, career-minded woman who didn't want to settle down. Now she just seems pathetic and always on the hunt for a man.
Is it definite that he meets her AT Barney's wedding, or he meets her BECAUSE of Barney's wedding? I know he mentioned the wedding to Wendy the Waitress in the future, but it wasn't 100% clear from what I remember. Maybe the Mother is the bass player in the band that's performing at the wedding, and she either offers Ted her umbrella (maybe after recognizing him as the lame professor who mistakenly taught her accounting class) or leave it behind at the wedding and somehow he gets it back to her, or Ted somehow puts two and two together that the umbrella's owner is Cindy's roommate that's supposedly perfect for him.
This. I also think Robin will marry Don because of the Uncle Don comment.

Yeah I don't think he ever said Uncle Don.
I think that Barney's half sister is a definite possibility. I have also heard a rumor that Marshall's mom ( she come to NY to help Marshall and Lily with the baby) and the Captain will get married and that the mother could also be the Captain's daughter. This will create a competition between Marshall and Barney to set Ted up with their respective sisters and be Ted's real bro.
But who knows. It could be someone completely different. We just have to see.
