Cover letter question

Some back story. I am a medical assistant. While taking a pt back to a room it came up that she was a nursing supervisor at a specialist office that happens to be closer to my home. She gave me a business card and told me to send her my resume. I had just started at my current job and wanted to wait it out to see how I liked it there and never sent her my resume.

Fast forward five months later my current office is not working out for me. I want to send my resume but don't know if I should send a regualr cover letter, or one adressed to the nursing supervisor jogging her memory about our discusion a few months back. Any advise would be greatly appreciated.
