I could watch these three movies on repeat and never grow tired of them In fact two of my favs are on FX today and I haven't gotten a single thing done cause I can't pry myself away.
1. Forgetting Sarah Marshall
2. Hangover
3. Step Brothers
Hmm. To answer this I have to limit it to recent comedies.

Monty Python would have to be my number one just because it's been in my life since I was a baby.
Probably a tie between Blazing Saddles and Young Frankenstein. Mel Brooks is the sh!t.

The Birdcage is fantastic.
I'd also like to add The Producers, Dumb and Dumber, and Ace Ventura 2. I can't explain the Ace Ventura one, except that it was the first DVD I bought and it cracks my sh!t up.
ETA: Roman Holiday is my all-time favorite movie, and I guess it's a comedy, but it's kind of bittersweet.
